Give reasons why :
(a) DNA cannot pass into a host cell through the cell membrane.

a) Since DNA is a hydrophilic molecule, it cannot pass through cell membranes. In order to force bacteria to take up the plasmid, the bacterial cells must first be made ‘competent’ to take up DNA.

b) Proteins can be removed by treatment with protease. So, protease is added to remove proteins when isolating DNA.

c) The presence of more than one recognition site within the vector will create several fragments, which complicate the gene cloning process. To avoid that the single cloning site is preferred in a vector.
