Four steps of preparing a temporary mount of leaf peel:
(1) Peel off the epidermis from the lower side of leaf by tearing it.
(2) place the peel in a glass containing water and then transfer the peel to water glass and add rafranin.
(3) Again transfer the peel back to glass to wash out excess stain. Lift the peel using needle for placing the peel on side.
(4) Pour a drop of glycerin on slide and place the peel on it and place a clean coverslip gently over it.
