When a gene of interest is inverted within the coding sequence \beta- galactosidase enzyme if the recipient cells fail to develop the blue colour in the presence of a chromogenic substrate. It is an indication of successful recombinant formation and present of the blue colour show no recombinants formed, whereas, antibiotic-resistant selectable markers used, as it requires simultaneously planting of two different antibiotics to differentiate between transformants from non-transformants to identify the recombinants from them, which is a recombinant from them. It is a very time consuming and cumbersome porous in comparison to the first one.
