Describe the process of amplification of "gene of interest" using PCR technique.
PCR involves three steps:
1. Denaturation:- The double-stranded DNA is heated to separate the two strands from each other(temp- )
2. Annealing:- The primers (small chemically synthesized oligonucleotides that are complementary to the region of the DNA (gene of interest) attach to the template DNA.(temp- )
3. Extension:- The enzyme DNA polymerase extends the primers using the oligonucleotides provided in the reaction and the genomic DNA as a template (temp-)
The process of replication is repeated many times so that DNA is amplified such repeated amplification is achieved by the use of a thermostable DNA polymerase i.e. Taq Polymerase isolated from bacterium Thermus aquanticus which remains active during high temperature.
