Option 4  is correct because the cytochemical marker acid phosphatase acts on the Golgi-associated endoplasmic reticulum lysosome (GERL). Acid phosphatase activities exist in GERL region and in side-cisternae which are mature and also in vacuoles which are condensing.GERL was examined with cytochemical markers, two enzyme markers (TPPase) and (ACPase) and osmium impregnation in the ameloblasts.
