a) 

  • Proinsulin has peptide chain A and chain B along with peptide chain C in the middle.
  • Mature insulin has only peptide chain A and chain B linked together by a disulphide bond 

b) They prepared two DNA sequences corresponding to chains A and B of human insulin and introduced them into the plasmids of E.coli to produce insulin chains.

Then, A and B were produced separately, then extracted and combined creating disulphide bonds to form human insulin.
