They grew E.coli, in ^{15}NH_{4}Cl. For many generations to get ^{15}N incorporated into DNA, then the cells transformed into ^{14}NH_{4}Cl, the extracted DNA is centrifuged in CsCl and measured to get their densities, DNA extracted from the after one generation (20 minutes), showed intermediate hybrid density, DNA extracted after two generations (40 minutes ) showed light DNA, hybrid DNA.
