a) rop- rop stands for “regulator of origin of plasmid”. This site is involved in the regulation of copy of the number of pBR322.

b) ori- ori stands for origin of replication. It is the site where DNA replication begins which enables the plasmid to reproduce itself to survive within the cell.

c) selectable marker- In addition to ‘ori’, the vector requires a selectable marker, which helps in identifying and eliminating non-transformants and selectively permitting the growth of the transformants.
