Protein found in biological system with unique 3D structure and biological activity is called native protein.

During denaturation, native protein may undergo physical change and chemical change. 

Due to change in temperature and pH, the hydrogen bonds in the protein are disturbed and globules gets unfold and helix starts to uncoil. Finally, the protein will lose its biological activity.
