The vector and the target DNA fragment can be sepeartely digested with the same restriction enzyme. The digested vector and the target DNA fragment are then incubated together in the presence of DNA ligase enzyme. Incubation results in bonding two types of DNA by phosphodiester bonds between them. Thus forming a recombinant DNA molecule.
