Occurs in the lumen of thylakoid. Requires Mn+, Cl-, Ca2+ and oxygen evolving complex

2H_{2}O\rightarrow 4H^{+}+4e^{-}+O_{2}

e- move towards P680 of PSII and H+ accumulate in lumen and finally reduced to NADP
