What modification is done on the Ti plasmid of Agrobacterium tumefaciens to convert it into a cloning vector?

Answers (1)

Ti plasmid in Agrobacterium can induce tumours in plants. This plasmid is 'disarmed' by suitable modification. A specific site of the T-DNA segment of the Ti plasmid is cut by a restriction enzyme and the DNA fragment with the gene of interest will be inserted in this segment using a ligase enzyme. Now modified, the T-DNA  will not induce tumours, however, the desired gene gets transferred into the host plant cell.
